Master regulator protein brings plethora of coactivators to gene expression sites

Molecular geneticists call big boss proteins that switch on broad developmental or metabolic programs "master regulators," as in master regulators of muscle development or fat metabolism. One such factor, the Activating Transcription Factor 6? (ATF6?) protein, takes charge following a cellular crisis known as endoplasmic reticulum (ER) stress, which is triggered by the accumulation of misfolded and aggregated proteins.

Molecularly, the ER stress pathway is always poised for action. Inactive ATF6? is normally embedded in cellular membranes, but at the first hint of protein overload, its working end springs superman-like into the nucleus, binds DNA and kicks on a host of target genes whose job is to clear a protein logjam.

Now, in a study published in the June 29 issue of The Journal of Biological Chemistry, and selected as "Paper of the Week" by the journal's editors, a team led by Stowers investigators Ron and Joan Conaway reveal that unlike the real superman ATF6? does not work solo. Using the ATF6? target gene HSPA5 as a probe, they apply mass spectrometry analysis to show that ATF6? recruits a fleet of coactivators to assist in target activation.

"We knew that as a master regulator, ATF6? was needed to turn on downstream genes in the ER stress response," says Ron Conaway, Ph.D., who with Joan Conaway, Ph.D., is co-corresponding author of the study. "Our goal was to determine what ATF6? was bringing with it to these genes' control elements."

"By devising a clever mix of state-of-the-art mass spectrometry and good old-fashioned biochemistry, this study has revealed that ATF6? is a virtual magnet for a wide range of 'A-list' co-regulators," said Michael K. Reddy, Ph.D., who oversees transcription mechanism grants at the National Institutes of Health's National Institute of General Medical Sciences, which partly supported the work. "These co-regulators offer a large array of proteins to target in efforts to control the ER stress response and to treat diseases that result from misfolded proteins."

That task of identifying co-regulators was challenging: labor-intensive molecular techniques the group applied to identify candidate interactors early on were not sensitive enough. At that point, the Conaways turned to their frequent collaborators Proteomics' Center director, Michael Washburn, Ph.D., and Laurence Florens, Ph.D., who heads the Stowers proteomics cores. Both had helped develop a sensitive mass spectrometry approach that can detect protein-protein interactions in highly complex mixtures, a technology known as MudPIT.

The group then set up a test-tube comparison. They genetically engineered a strand of DNA flanking the HSPA5 target gene, the so-called "enhancer" region recognized by ATF6?. They then dipped two identical DNA test strips into respective pots of cellular extracts -- one containing ATF6? and one not -- reasoning that factors in the ATF6? entourage would be recruited to the first but not the second. They then applied a single run of MudPIT to identify each ATF6?-specific partner.

In short, they found that it takes not a village but a metropolis to activate an ATF6? target. Many proteins bound the enhancer in both samples, meaning either that they're just background, or else that they must bind DNA even when the gene is inactive. But more than 40 were present in about 5-fold excess only in ATF6??spiked samples, suggesting they are tethered to the enhancer by ATF6?.

Among the latter were components of a multi-subunit behemoth protein known as Mediator, which bridges specific genetic switches (like ATF6?) and the catalytic machinery that copies a gene. Other proteins recruited by ATF6? through overlapping but not identical domains belonged to other large complexes known as SAGA and ATAC, which enzymatically relax chromosome structure to allow gene expression.

Researchers know that all DNA-binding factors partner with other proteins to switch genes on or off. What is remarkable here is their sheer number. "It would be very interesting to find out whether this is the norm," says Ron Conaway. "This work raises a ton of little questions about mechanism."

Among them is how do ATF6?-interacting factors arrange themselves on the test strip, and does a single ATF6? bind to all of them at once? "There are three separate ATF6? binding sites on the HSPA5 enhancer and ATF6? itself forms a dimer," explains Dotan Sela, Ph.D., a Conaway lab postdoc and the study's first author, "So potentially within this region there could be as many six activation domains," he explains.

Solving these puzzles could reveal molecular targets for seemingly unrelated diseases. While a little ATF6? signaling is absolutely essential for cellular housekeeping, unrelieved ER stress is a hallmark of neurodegenerative conditions like Alzheimer's and Huntington's Diseases and is correlated with insulin insensitivity and type II diabetes.

A direct role for ATF6? in what some now call "misfolded protein diseases" is unclear. Nonetheless, the study suggests ways to dampen ER stress signaling molecularly. "We show that the Mediator is relevant to HSPA5 expression," says Sela. "So one way to keep ATF6? from turning on a gene might be to devise ways to block binding of the Mediator to ATF6?."

Joan Conaway also points out that MudPIT data analysis does not require previous identification of a "suspect." "Our approach complements methods that test candidate interactors one by one," says Joan Conaway. "Because the analysis is unbiased, it could reveal novel proteins interacting with a particular enhancer, which then could be confirmed using other methods."

The Conaways began their pioneering studies of mammalian gene expression over three decades ago, when only laborious biochemical techniques were available. As a result, both deeply appreciate what a technological leap the current work represents. "This study provides proof of principle for the utility of mass spectrometry in defining novel transcriptional activators," says Ron Conaway. "We want to compare this data with that from other activators -- it's what we will be working on in the future."

In addition to Washburn and Florens, Lu Chen of the Conaway lab and Skylar Martin-Brown of the Washburn lab also contributed to the work.

Chemotherapy: tips for managing side effects | Cancer Council

The most important effect of chemotherapy is that it kills cancer cells. However, chemotherapy can also affect normal cells that rapidly divide and grow, such as those in the bone marrow, digestive tract, skin, hair and reproductive organs. When the normal cells are damaged, this can cause side effects.

This chapter discusses the most common side effects people experience and offers suggestions to help manage them.

  • It is natural to worry about side effects. However, whether or not you experience side effects, and how severe they are, depends on the type of drugs you?ve been prescribed and how you react individually from one treatment period to the next.
  • Side effects vary greatly. Some people will have no side effects, others will experience a range. If you have side effects, they will usually start during the first few weeks of treatment.
  • Most side effects are temporary and can be managed. They tend to gradually disappear once treatment has stopped.

You should talk to your doctor or nurse before your treatment begins about the side effects you should watch out for or report, and who to contact out of hours if you have immediate concerns.

Chemotherapy diary

Share the information you record with your doctors and nurses so that they can give you suggestions for dealing with side effects or adjust your treatment, if appropriate.

Feeling tired and lacking energy (fatigue) is the most common ? and often, debilitating ? side effect of chemotherapy. Fatigue can include feeling exhausted, drowsy, confused or impatient. You may have a heavy feeling in your limbs, or find it difficult to do daily activities.

Fatigue can appear suddenly and rest may not relieve it. You might still feel tired for weeks or months after a treatment cycle ends.

  • Save your energy. Help your body recover by doing less and resting more.
  • Let people help you. Family, friends and neighbours often want to assist but may feel unsure about what to do. They could help with shopping, child-care, driving or housework.
  • If you are employed, consider whether you could take some time off, reduce your hours or work from home during or after chemotherapy. Some workplaces might allow you to work flexibly around your treatment.
  • If there is a time during the day when you tend to feel most energetic, try to plan activities during this time.
  • Do light exercise, such as walking. If you have a normal exercise routine and you feel up to it, ask your doctor if you can exercise.
  • Don?t start any new exercise activities until you have recovered from treatment.
  • Try to get more sleep at night and take naps during the day if you can.
  • If you aren?t sleeping well, tell your health care team. Only take sleeping pills that are recommended or prescribed by your doctor.
  • Some people benefit from doing relaxation or meditation exercises.
  • Try to eat a healthy, well- balanced diet with plenty of fruits and vegetables.

  • Check with your doctor whether your fatigue is related to low red blood cells (anaemia) so that this can be treated.

It is common for your appetite to change when you are going through chemotherapy. Sometimes you may not feel hungry, or you may not enjoy the foods you used to like. Some treatments temporarily change the taste of foods.

Chemotherapy may make you feel sick (nauseous) or vomit. Not everyone feels sick during or after chemotherapy, but if nausea affects you, it usually starts a few hours after treatment. Nausea may last for many hours and be accompanied by vomiting or retching.

Anti-nausea or anti-vomiting medication can help. This medicine can be taken before, during or after treatment. It may be available as:

  • injections ??usually given before chemotherapy
  • tablets ??can be taken regularly at home
  • wafers ??dissolved under or on top of the tongue
  • suppositories ??placed in the rectum where they dissolve
  • liquids ??added to the chemotherapy.

Your medical oncologist will tell you if the drugs you are given are likely to cause nausea and vomiting. You may be prescribed medication in advance so you don?t experience the side effects, or so they are reduced. It may take some time before you find a medication that works for you, so let your nurse or doctor know?if you still have nausea after a few days taking the medication or if you have vomited for more than 24 hours.

Trying to eat as well as possible will help you recover sooner and cope better with the treatment and its side effects.

Tips

  • Eat a light meal before your treatment (e.g. soup and dry biscuits or toast), and drink as much fluid as possible.
  • Sip small amounts of liquid often, rather than trying to drink a lot at once. Sucking on ice cubes, ice-blocks ?or jellies can also help to increase your fluid intake.
  • If your stomach is upset, try drinking fizzy drinks such as soda water or dry ginger ale.
  • If you wake up feeling sick, eat a dry biscuit or a slice of toast rather than skipping food altogether or forcing yourself to have a full meal.
  • Breathe deeply and gently through your mouth if you feel like you?re going to vomit.
  • If you are nauseous or have vomited a lot, try to keep your fluids up so that you don?t get dehydrated. If you aren?t able to keep fluids down, contact your doctor immediately.
  • Avoid strong odours and cooking smells.
  • Eat and drink slowly. Chew your food well to make it easier to digest.
  • Prepare meals between treatments and freeze them for the days you don?t feel like cooking.
  • Be flexible about your meals. Try to eat when you are hungry, and don?t delay eating when you feel like it.
  • Eat small, frequent snacks instead of large meals.
  • If the taste of certain types of food has changed, don?t force yourself to eat them. Your sense of taste should return to normal after treatment ends, but it may take several weeks.
  • Speak to the hospital dietitian for advice about eating.
  • Call Cancer Council Helpline 13 11 20 for free information about eating healthily during and after treatment.

Many people having chemotherapy worry about hair loss. Some people lose all their hair quickly, others lose it after several treatments, and others may only lose a little hair or none at all.

  • Whether or not you lose your hair depends on the drugs you receive. Some drugs don?t cause this side effect. Ask your doctor if this is a possible side effect and discuss how you can prepare for it.
  • When hair loss does occur, it usually starts 2?3 weeks after the first treatment and grows back when chemotherapy is completed. Your scalp may feel hot, itchy, tender or tingly before and while your hair is falling out. Some people find that the skin on their head is very sensitive, and they may develop pimples on their scalp.
  • Although losing head hair is most common, you may also lose hair from your eyebrows, eyelashes, arms, legs, chest and pubic region.
  • It takes 4?12 months to grow back a full head of hair. When your hair first grows back, it may be a different colour or curly (even if you have always had straight hair). In time your hair usually returns to its normal condition.
  • Many people find losing their hair very difficult. You may feel that hair makes up a part of your overall image and its loss can make you feel physically unattractive, vulnerable or sad. It?s natural to feel this way. Talking to your medical team may be helpful.

Tips

  • Keep your hair and scalp very clean.
  • Use a mild shampoo like baby shampoo. If you want to use lotion on your head, use sorbolene. Check with your nurse before using any other hair or skin care products.
  • Comb or brush your hair gently using a large comb or hairbrush with soft bristles.
  • Wear a light cotton turban or beanie to bed if you are cold at night, or to collect hair.
  • Use a cotton, polyester or satin pillowcase, as nylon can irritate your scalp.
  • If you prefer to leave your head bare, protect it against sunburn and the cold.
  • Make an appointment? with a hairdresser who can make your hair look as good as possible even if it is thin or patchy.
  • Avoid perms and dyes, and limit the use of hair dryers, rollers and harsh products.
  • If your eyelashes fall out, wear glasses or sunglasses to protect your eyes from the sun and dust while outside.
  • Tell your nurse or doctor if the skin on your scalp is very sensitive, or if you have a lot of discomfort ?and itchiness.
  • Wear a wig, toupee, hat, scarf or turban. Do whatever feels the most comfortable and gives you the most confidence.
  • Some hospitals or treatment centres have a wig library where you can borrow a wig if you need one. Ask your doctor or nurse for more information.
  • If you want to purchase a wig and you have private health insurance, check with your fund to see if you are eligible for a rebate.
  • Call Cancer Council Helpline 13 11 20 for assistance in finding a wig library or shop.

Your skin may darken, peel or become dry and itchy when you are having chemotherapy. It is also likely to be more sensitive to the sun. Protect your skin from the sun ? especially between 10am and 3pm ? by wearing high-protection sunscreen (SPF 30+), a hat and protective clothing. This advice applies to everyone, but it is even more important for people having chemotherapy.

Tips

  • Shower using a moisturising soap or sorbolene cream as a soap replacement.
  • After showering, gently pat your skin dry with a towel. Try not to rub your skin too hard.
  • Use a moisturising lotion or cream containing the ingredient urea to stop the dryness.
  • Dust cornflour over the itchy parts of your skin.
  • Wear loose, non-restricting clothing. Choose cotton fabric instead of rough wool or synthetic fibres.
  • Wash your clothing in mild detergent for people with sensitive skin.
  • Stop shaving or waxing until your skin is completely healed.
  • If your skin becomes red or sore in the area where the intravenous device went in, tell your doctor or nurse immediately.

Some chemotherapy drugs, pain relief medicines and anti-nausea drugs can cause constipation or diarrhoea. Tell your doctor or nurse if your bowel habits have changed.

After a bout of constipation or diarrhoea, eat a balanced diet with fresh fruits, vegetables, wholegrain bread and pasta.

Constipation

?

  • Eat more high-fibre foods, such as wholegrain bread and pasta, bran, fruit and vegetables, nuts and legumes (e.g. baked beans or lentils).
  • Sometimes drinking fruit juice can help ease constipation. Try prune, apple or pear juice.
  • Drink plenty of other fluids, both warm and cold, to help loosen the bowels.
  • Do some light exercise, such as walking.
  • Don?t use enemas or suppositories. They are not recommended for people having chemotherapy.
  • If you have constipation for more than a couple of days, let your doctor or nurse know. They may change your medication or give you other medication to relieve it.

Diarrhoea

  • Choose bland foods such as clear broth or boiled rice. Avoid spicy foods, wholegrain products, fatty or fried foods, rich gravies and sauces, and raw fruits or vegetables with skins or seeds.
  • Limit alcohol, fruit juice, soft drinks, strong tea or coffee and dairy products, as these may further stimulate the bowel.
  • Drink water to help replace the fluids lost through diarrhoea.
  • Diarrhoea can usually be treated at home with over-the-counter medication. Talk to your doctor or pharmacist.
  • If the diarrhoea becomes severe, it can cause dehydration and you may need to be admitted to hospital.

Some chemotherapy drugs can cause mouth sores such as ulcers or infections. This is more likely if you have had or are having radiation to the head, neck or chest, or if you have dental or gum problems.

If you notice any change in your mouth or throat, such as sores, ulcers or thickened saliva, or if you find it difficult to swallow, contact your doctor. You should also discuss any dental problems with your doctor before seeing the dentist. If you need any dental work, tell your dentist you are having chemotherapy.

Tips

  • Use a soft toothbrush to clean your teeth twice a day.
  • Don?t use commercial mouthwashes containing alcohol as they may dry out or irritate your mouth. Instead, try homemade mouthwash (1 tsp bicarbonate of soda or salt in a glass of warm water) at least four times a day. This helps keep your mouth clean, while the salt mouthwash helps heal mouth sores.
  • Sip fluids, especially water, and eat moist foods such as casseroles or soups if you have a dry mouth. Moisten foods with butter and sauces.
  • Try sucking on ice while you?re having intravenous chemotherapy to reduce mouth ulcers.
  • Blend foods to make them easier to eat.
  • Nourishing drinks such as milkshakes, smoothies and supplements add extra energy to your diet and feel soothing.
  • Avoid very hot foods, and spicy, acidic or coarse foods, (e.g. nuts or grains), as these can aggravate mouth sores.
  • Don?t smoke or drink alcohol, as this irritates tissue in the mouth.

Some people say they have trouble thinking clearly after chemotherapy. This is called cognitive impairment or, sometimes, ?chemo brain?. You may find that it takes you more time to process information, or you may experience short-term memory loss or have trouble concentrating for long periods.

There is some evidence that self-help techniques such as relaxation, exercise and memory games can be beneficial. You can also try focusing on activities that don?t require too much concentration. For example, you might choose to read magazines with short articles instead of books with several characters or complex storylines. Tell your doctor if this issue is affecting your day-to-day life.

Tips?

  • Use a calendar to keep track of tasks, appointments, social commitments, birthdays, etc.
  • Write down things you have to remember. For example, make to-do lists or write down where you parked the car.
  • Learn something new, take up a new hobby, or do crosswords, ?word or number puzzles.
  • Plan activities to do things that require more concentration when you are fresher, such as in the morning.
  • Get plenty of sleep. Deep sleep is important for memory and concentration.
  • Do light exercise each day as this may help you to clear your mind and sleep better.
  • Talk to others about how you?re feeling ? for example, your partner or colleagues ? to prevent misunderstandings and frustration.

Some chemotherapy drugs affect the bone marrow, which is the soft and spongy material inside the bones. The bone marrow makes three types of blood cells:

  • red blood cells ??carry oxygen throughout the body
  • white blood cells ??fight infection
  • platelets ??help blood to clot and prevent bruising.

The bone marrow?s job is to maintain normal levels of blood cells (your blood count) to keep you fit and healthy. When chemotherapy affects the bone marrow, your blood count is reduced. The count may fall with each treatment. This can cause problems such as anaemia, infections and bleeding problems, depending on the type of blood cell affected.

Your health care team will do regular blood tests to make sure your blood cells return to normal levels before your next treatment.

Anaemia

  • A low red blood cell count is called anaemia.
  • This can make you feel tired, lethargic, dizzy or breathless.
  • You may need a blood transfusion to build up your red blood cells and treat the anaemia.
  • Eating a nourishing diet with foods rich in iron and B vitamins is also important if you are anaemic. Wholegrain breads and cereals, lean meat and green leafy vegetables are good sources of iron and vitamin B.

Infections

If white blood cell numbers drop during chemotherapy treatment, it can make colds and flu harder to shake off. Scratches and cuts may get infected easily or take longer to heal.

See your doctor if you are unwell ? even if you just have a cold. Sometimes doctors recommend taking antibiotics as a precaution against infection. If you are having chemotherapy in winter, check with your doctor about having the flu vaccine.

You may have an injection of granulocyte-colony stimulating factor (G-CSF) after chemotherapy if your doctor thinks it will be useful for you. It increases the number of neutrophils, which are a type of white blood cell that help protect against infection. If G-CSF is given, your doctor or nurse will speak to you about possible side effects. Some people may experience bone pain or tenderness at the injection site or show signs of an allergic reaction.

When to contact your doctor

Contact your doctor urgently if any of the following occur:

  • chills or sweats
  • a temperature of 38?C or more
  • persistent or severe vomiting
  • severe abdominal pain, constipation or diarrhoea
  • unusual bleeding
  • tenderness, redness or swelling around the site of the intravenous chemotherapy device
  • any serious unexpected side effects or sudden deterioration in health.

Also, contact your GP or treating specialist if you experience:

  • sweating, especially at night
  • sore throat
  • mouth ulcers
  • burning or stinging on passing urine
  • easy bruising.

Tips

  • Keep a thermometer at home to check your temperature.
  • Some people having chemotherapy prefer to avoid crowded places such as shopping centres or public pools to reduce the risk of picking up an infection. This is not always practical, so use your commonsense.
  • Wash your hands with soap and water before preparing food and eating, and after using the toilet.
  • Use separate boards and utensils for raw and cooked foods, and clean thoroughly with hot, soapy water.
  • Leftover food should be stored in airtight containers in the fridge as soon as possible.
  • Store raw and cooked foods separately.
  • Try to eat freshly cooked or freshly prepared foods.
  • Don?t eat food or drinks past their use-by or best before dates.
  • Wash fruits and vegetables well, or peel where possible.
  • Avoid pre-made sandwiches, salads and takeaway foods; soft cheeses; raw or rare fish, meat and eggs; unpasteurised dairy products and soft-serve ice-creams.
  • Avoid people who are unwell, especially if they have the flu, conjunctivitis, measles, mumps, a cold sore or chickenpox.

Bleeding problems

A decrease in platelets can cause you to bleed for longer than normal after minor cuts or scrapes, or to bruise more easily. You may need a platelet transfusion if they are low.

Tips

  • Be careful when using scissors, needles, knives or razors. Small cuts or nicks can harbour germs where an infection can start.
  • Use an electric razor to minimise the chance of nicking yourself.
  • Use a soft toothbrush to avoid irritating your gums.
  • Wear thick gloves when gardening to avoid injury, and to prevent infection from soil, which contains bacteria.
  • If you bleed, apply pressure for about 10 minutes and bandage, if appropriate.

Some drugs can cause tingling and loss of sensation in your fingers and/or toes, and muscle weakness in your legs. If this happens, tell your doctor or nurse before your next treatment.

Your treatment may need to be changed or the problem carefully monitored.

Marriage Communication: Understanding the Importance | Pursuit of ...

Here is an article I wrote for His N Her Love Story Blog a few weeks back. Thought I would share with you for today?s Marriage Monday.

When you meet a couple that has been married for a long time and you ask them what their secret to a successful marriage is: more than likely they will tell you that communication was at the top of the list. Communication is seen as the doorway to understanding therefore should be used wisely in our marriage relationships. Communication is the key to a strong and healthy marriage.

For example, as a wife, we often complain about our husbands not understanding what we are feeling at the time yet more often than not we do not effectively communicate with our spouses. First we have to learn that women and men are created differently, therefore the waves of communication is different.

Communication is the only way you will know what is going on and how your spouse feels when it comes to things in your marriage. You need to talk to your spouse about how you feel about things they do or don?t do. A marriage is built on communication and you won?t have that successful marriage if you are not communicating with each other effectively.

You may think, ?Well, we have love for each other,? in which that is good but what is love when you have no way of nurturing the love you have for each other through communication? Here are a few tips that will help you understand the purpose and importance of communication in your marriage:

  • Identification ? Letting your spouse know you accept them as is.
  • Significance ? Letting your spouse know that they are the most important person in your life.
  • Considerations ? Letting your spouse know that you are thoughtful, attentive, considerate and careful of their needs.

Free Workplace ESL program enhances workplace communication ...

Milton, ON: Last week, employees at Kenco Logistic Services celebrated a special milestone in their careers?graduation from the Workplace ESL (English as a Second Language) program. Over the past 10 weeks, nine employees participated in classes designed to help them better communicate in English to meet the needs of their specific workplace. The program is funded by the Ontario Ministry of Citizenship and Immigration and coordinated by The Centre for Skills Development & Training.

?

?Kenco is a forward-thinking company that recognizes the economic benefits of hiring skilled newcomers,? said Kathy Mills, The Centre?s CAO. ?They also recognize how important English literacy is to today?s workplace. By participating in the Workplace ESL program, they have demonstrated a willingness to invest in their workforce?and in their community.?

?

Kenco?s Whirlpool Regional Distribution Centre is a state of the art facility that employs more than 100 people. Many of the company?s workforce are new Canadians, and, while they are highly skilled and trained in completing their job duties, some still struggle when it comes to communicating clearly in English. Kenco is the first company in Milton to take advantage of this specialized language training program, which is free to employers and employees.

?

?Our Workplace ESL program is designed to help a company?s employees?particularly those who are new to Canada?become more proficient in their jobs, learn specific job functions, and prepare for more responsibility,? said Deborah Dennison, Coordinator of the program. ?At the end of the program, employees have more confidence, can work more safely, communicate more effectively, and better represent the company.?

?

Many of the employees who participated in the program already had a high level of English language reading, writing and speaking skills, and all were university educated. The Centre worked closely with Kenco to develop a customized curriculum that included lessons on language specific to their work environment and on health and safety processes.

?

?Employees who took the training are more confident, and they have improved spelling, grammar and pronunciation, which helps with problem-solving and teamwork,? said Gloria Chaumont, HR Manager at Kenco. ?We have also seen improvement in their understanding of health and safety policies and procedures, and knowledge of acronyms that are common in today?s business world.?

?The classes were not only great learning experiences; everyone had fun!? continued Ms. Chaumont. ?There was always a lot of laughter coming from our training room when the classes were in session, which added to the team-building experience. We thank the folks at The Centre for bringing this to our company.?

Man sentenced to life in Michigan stabbing spree

(AP) ? A man convicted of murder in a 2010 stabbing spree in Michigan was sentenced to life in prison Monday, wrapping up the first in a series of cases against Elias Abuelazam.

Michigan offers no parole to people convicted of first-degree murder, which means the 35-year-old Israeli immigrant will be locked up until he dies, unless he wins an appeal.

The first of 10 possible trials stemming from the stabbing spree centered on the death of Arnold Minor, a 49-year-old handyman who was attacked in the early hours of Aug. 2, 2010, near downtown Flint. The evidence was virtually impossible to overcome: The victim's blood was found in Abuelazam's Chevy Blazer and inside his luggage.

Before the mandatory sentence was announced, Minor's mother, Elzora Minor, collapsed and was carried out of court. She had been standing in front of the judge holding her son's cremated remains, alongside daughter Stephanie Ward, who had a biting farewell for Abuelazam.

"I'm just glad for this day and hope that he rots in hell for the pain he put this city through," Ward told Genesee County Judge Judith Fullerton.

Outside court, Elzora Minor, 68, said she felt overwhelmed and fell when she looked over at Abuelazam and saw him grin.

Abuelazam was given a chance to speak but said nothing about the crime. Fullerton, unlike many judges in high-profile cases, didn't offer any tough words before the convicted killer was taken away in chains.

Defense lawyers said Abuelazam was paranoid schizophrenic and under the spell of demons that forced him to stab and kill. But three experts hired by prosecutors interviewed him and found he wasn't mentally ill or incapable of understanding the criminal acts.

The stabbing spree terrified the working-class city of Flint. Fourteen people were stabbed in the area, and five died, including Minor. Survivors said their assailant claimed to have car trouble or asked for directions before attacking.

Most were stabbed in the wee hours. There is no evidence that Abuelazam, who worked the afternoon shift in a party store, knew them. A motive has not been offered.

"This man terrorized the community. He murdered our citizens. ... He stabbed and maimed our friends," Genesee County Prosecutor David Leyton told the judge Monday.

Abuelazam had lived in Flint for just a few months, staying in a house owned by his uncle, who lived next door. He was captured in Atlanta in August 2010 while trying to flee to Israel.

He faces two more murder trials and six attempted murder trials in Michigan as well as an attempted murder case in Toledo, Ohio. It's not clear, however, whether prosecutors actually will pursue that many trials.

With a life sentence now secured, Leyton said he's open to plea deals but first wants to talk to victims and relatives of victims. The judge won't hold another court hearing until late summer.

"I have to balance their feelings with my fiduciary duties to the county," Leyton told The Associated Press, referring to the cost of so many trials.

Microsoft Surface tablet may cost $599, have Wi-Fi only

Sources cited by TheNextWeb say that the base price for a?Surface tablet will be $599 ? and the Intel-powered "Pro"?version will start at $999. Meanwhile, Bloomberg is reporting that the initial models will be Wi-Fi-only, rather than connecting to a wireless 3G or 4G network.

I reached out to Microsoft, but the company was unable to confirm either of the rumors. However, it's worth looking at why both are relatively likely.

(Msnbc.com is a joint venture of Microsoft and NBC Universal, but that doesn't give us privileged information about Microsoft products and services, nor does it influence our opinion of them.)

First, the pricing. At the Surface unveiling, Microsoft announced that the tablets would be "priced competitively" with current tablet offerings, and that the ARM-based Surface, meant to compete with the iPad, would ship in 32GB and 64GB versions. Well, a 32GB Wi-Fi-only iPad costs, yes, $599. So that's definitely in line.?

But Microsoft said "competitive," and if Microsoft really?wants to compete with Apple, it probably has to undercut the iPad. So while the $599 is more than likely, it would be nice to see Microsoft enter the market aggressively, rather than launch high, then drop the price if (when?) sales are slow. Amazon didn't price its Kindle Fire high then drop it, it went on the cheap. Meanwhile, RIM priced its PlayBook very high, then dropped and dropped and dropped the price. Which company is holding its head high now?

As for a $999 Pro version, that's a bit more of a puzzlement. All Microsoft has said was that the pricing would be in line with current ultrabooks, but quick survey of current ultrabook pricing suggests a starting point of $699 or so. Admittedly, that's without a 64GB solid state drive, let alone 128GB, which the high-end Surface Pro will have, but these ultrabooks do have larger screens. So while $999 may not be a bad price for what Microsoft is offering, it seems a little high to compete with ultrabooks in general.

The Wi-Fi-only issue is far easier to wrap your head around. At the Surface event, Microsoft showed off only hardware that ran on Wi-Fi networks -- they said absolutely nothing about cellular 3G/4G?networking. They also announced that they'd have the basic Surface tablet out when Windows 8 launches, and the Pro version out?about?three months later. By showing off Wi-Fi-only?hardware and talking about its availability, they've all but formally confirmed that there won't be a cellular product "initially," which is all Bloomberg is saying.

So there you have it. If you're in the market for a Surface tablet,?don't expect a 3G/4G version at launch, and do?expect to pay around $599 for the entry-level unit. And if the price happens to be lower, that's money in your pocket.

Surviving Student Loans

For the next week (or two), we?ll be sharing ?audition? pieces from folks interested in being new staff writers at Get Rich Slowly. Your job is to let us know what you think of each of these writers. The student loan juggernaut, before it became a national scandal, was a way of life for many middle-class high school students like myself. Better known as a ?necessary evil,? than a reason to be embarrassed or worried.

But you can?t change the past, so here is advice for students past and present ? whether you?re about to make the leap and want a glimpse of your future or you are still recovering from the fall.

Prep for Success
Student loans do not equal a free education, even if it seems like it at the time you sign on the dotted line. Put the math in terms you can accept: According to its 2012-2013 rates, one year at Princeton is like buying 109 iPads and paying for them for the next 30 years. Four years at Penn State is the same as buying a 1,867-year subscription to Forbes magazine. Talk about a legacy.

Many high schoolers just don?t think about what student loan debt means (not to discredit those who do, of course). But there?s a fine line between heading off to college because it?s what will benefit you the most and just doing what all of your classmates and peers are doing.

And remember: People will actually pay you to go to school. In the very rare case, a company you work for will foot the bill in exchange for years of your service. In other circumstances, nonprofits have money they need to get off their hands. They want to give it to you. You just have to prove that you aren?t a scammer and that you deserve it by responding to any and every essay scholarship you can find.

Minimizing Damage While Attending
It seems like a common phenomenon for today?s college student to see attending college as a social outlet and not the investment in one?s future it really is. Newsflash: College ends. Friends move out of town, and the money stops coming in unless you get a competitive job.

By all means make the most of the social aspects of college, because networking is valuable too, but don?t over-network at the expense (literally) of your future financial happiness. Move off-campus and get your hands on some sub-$500 rent. Pick up an hourly job at Blockbusters (oh wait, they?re closed) or a super-sketchy gas station (oh wait, I regret that) and put that money not towards the bar tab, but towards your groceries. Then cook and pre-game at home to keep restaurant and alcohol costs down.

The very last thing you should be doing is rounding up when you calculate your expenses and using the ?leftovers? as padding. Student loans don?t exist to make your life easier, they exist to help you get an education. Don?t take advantage because even if you skimp you?ll be paying for a long time.

Most importantly, major in something that will make money and leave your passions for your minor.
You can read Hemingway and ponder 1700s French philosophy on your commute home from a high-paying corporate gig. You can even save for a few years while working there and then quit to do what you want while you pay your own way. The alternative is having some really cool ideas and then trying to make it in the freelancing world right out of college. The alternative isn?t that great.

Consolidating and Paying Off
Maybe all of this information comes too late. Well, never fear, the US government is here.

If you have loans spread among different banks and all of the loans are in your name, you may be eligible to consolidate those loans into one big loan (and one big payment) by visiting Direct Consolidation Loan, a government website.

You?ll also have the option of choosing an Income-Based Repayment plan if you aren?t making very much, or even getting your loans forgiven after a period of time for working in a public service industry (check the website for more details).

Stay Motivated

Sandusky accuser: Wrestling led to sexual abuse

A 30-year-old Ohio man is the first accuser of former Penn State football Coach Jerry Sandusky to speak publicly about the sexual abuse he says he endured.

Travis Weaver, in an exclusive interview to be broadcast Thursday, June 21 at 10pm/9c on?NBC's Rock Center, said Sandusky performed oral sex on him in the upstairs bedroom of the Sandusky home, right across the hall from Sandusky?s wife, Dottie.

Weaver said Dottie Sandusky?never witnessed firsthand any of the abuse but he suspects she had an idea of what was going on.

?How could you not know?? asked Weaver in the interview.

The trial has grabbed headlines for the past two weeks. Though graphic details from the testimonies of eight of his accusers have been reported, none of the alleged victims of Sandusky has spoken publicly until now.

Weaver has testified in front of a grand jury but was not called as a witness in the current trial. He said he?is prepared to testify in the future if needed. He has spoken to both state and federal authorities.

Sandusky is facing 48 charges relating to child sex abuse. He has maintained his innocence. Attorneys gave their closing arguments June 21. At the time of this publishing, no verdict had yet?been reached.

Weaver said he had never told anyone of the abuse and had buried the memories deep down. He says he thought he was the only victim until he saw?reports that Sandusky?had been arrested on charges of molesting other boys last fall.

?I was shocked.? I couldn't believe he just kept doing it to all these other kids,? Weaver said.


Weaver said he feels guilty about the young men who said they were abused after him.

?I wish I would have said something to him.? I think if I had said something to him a lot of this stuff wouldn't 'a happened to all these other kids,? he said.

His attorney, Jeff Anderson, has represented dozens of victims of sexual abuse, many of whom were abused by Catholic priests.

?Speaking out is part of the process.? Travis is helping other children by telling his story. He is helping to protect other kids,? said Anderson.

Weaver said he met Sandusky in 1992 at the Penn State outdoor swimming pool when he was ten years old.? He was there with his younger brother as part of a Second Mile summer camp.

?It was like meeting my hero,? he said.

The sexual abuse began to occur gradually, he said, and started in the Penn State locker room.

?After the shower was over?he'd dry me off with a towel.? He'd say he was trying to wrestle with me....and then it progressed into oral sex,? he said.

Weaver said Sandusky abused him more than 100 times in the Penn State locker room, at the Sandusky home, and even in a hotel in Pasadena, California where Weaver and the Sandusky family stayed while on a trip to the Rose Bowl.?

?We went to a professional football game, and [Sandusky and I] left early and went back to the hotel.? And he performed oral sex on me in the hotel,? Weaver said.

Dottie Sandusky testified in court that her husband is innocent. She said she never witnessed inappropriate contact between her husband and young boys.

Weaver said that when he was 14, he moved away from the State College area to get away from Sandusky.

Weaver has filed a civil suit against Sandusky, Penn State and the Second Mile, alleging that the institutions could have done more to stop the abuse.

Sandusky?s attorneys could not be reached for immediate comment on this story.

A spokesman for Penn State declined to comment on this story.? In November 2011, the university?s Board of Trustees issued a statement saying it was outraged by the horrifying details contained in the Grand Jury report, which included the testimonies of eight accusers, aged 18 to 28.

?We cannot begin to express the combination of sorrow and anger that we feel about the allegations surrounding Jerry Sandusky. We hear those of you who feel betrayed and we want to assure all of you that the Board will take swift, decisive action,? the Board of Trustees said.

The Second Mile charity declared bankruptcy after losing significant funding in the wake of the Sandusky scandal. Second Mile Acting CEO David Woodle said his organization is continuing to cooperate fully with the Attorney General's investigation and will adhere to its legal responsibilities throughout this process.

The charity released a statement in November stating that to its knowledge, all the alleged incidents occurred outside of its programs and events.

?Our highest priority always has been and will continue to be the safety and well-being of the children participating in our programs. We encourage program participants to report any allegations of abuse and/or inappropriate sexual activity wherever it has occurred, and we take any such reports directly to Child Protective Services. We have many policies and procedures designed to protect our participants, including employee and volunteer background checks, training and supervision of our activities,? the statement said.

"Rock of Ages" succeeds on some levels and is simply silly on others.

The biggest surprise: this movie is funny (intentionally). Sure, there?s a trite love story, but the supporting cast provides quite a few laughs. What takes up most of the time is a string of anthem rock classics sung by every member of the cast. Some are fun and successful (the best gay love song ever), but many are downright embarrassing. Tom Cruise, as the metal god, is sometimes swaggering cool, sometimes funny. Adam Shankman?s direction/choreography is occasionally dazzling. Overall, this non-stop barrage of rock music videos succeeds on some levels and is simply silly on others.

GAY ANGLE: The gay love song is one of the film?s most inventive highlights.
